Affinity ITC

Isothermal titration calorimetry (ITC) is a highresolution, labelfree technique that measures the heat released or absorbed when molecules interact. It directly provides binding constants, reaction stoichiometry, enthalpy, and temperaturedependent behavior.

The Waters TA Instruments Affinity ITC combines an ultrasensitive calorimeter with a precision syringe that delivers controlled, minute injections into the calorimetric cell. Softwaredriven operation enables consistent, timed titrations, and optional automation allows sampling from temperaturecontrolled 96well plates for highproductivity, unattended workflows. Automated cleaning reduces carryover, and automated loading ensures reproducible sample delivery.

The Affinity ITC and ITC Auto are engineered for demanding lifescience environments requiring high sensitivity and advanced ITC performance. Together with the Nano DSC, the Affinity ITC supports genetherapy applications, including characterization of serotype and drug loading in AAVs and LNPs, as well as target and offtarget binding interactions. Its advanced engineering ensures the highestquality calorimetric data.

Because ITC requires no dyes or tags, researchers can work under their preferred conditions while preserving native molecular behavior. Stoichiometry is obtained during each experiment, enabling verification of 1:1 binding interactions without additional assays.

By delivering detailed affinity, specificity, and stoichiometry data in a single experiment, the Affinity ITC supports confident decisionmaking in drug discovery. NanoAnalyze software streamlines realtime data analysis, overlay generation, and rapid comparison of candidates.
